The most frequently used synthetic auxin - 2,4-D (2,4-dichlorophenoxy acetic acid) is necessary for callus formation and induction of somatic embryogenesis. The actual level of 2,4-D in the explant is an important factor for dedifferentiation or development of in vitro culture. The uptake and metabolism of 2,4-D in isolated cucumber cotyledons in vitro was studied in relation to different light conditions and growth-expansion of explants. The uptake of 2,4-D by cotyledons increases with time. Uptake of2,4-D by green autotrophic cotyle-dons was higher than by early heterotrophic cotyledons. During a 20-hour culture in light the cotyledons accu-mulated 2,4-D, green cotyledons more than hete-rotrophic cotyledons, in spite of the high intensity of 2,4-D metabolism in green cotyledons. The 2,4-D content and metabolites during 72 hours of culture changed.
