Rapidly developing methods based on molecular markers considerably contribute to the differentiation, characterisation and testing of individuals and their progeny. In our laboratory we mostly use RAPD for the differentiation of the individual clones oflong-term-cultivated somatic spruce embryos; lately we have introduced also the AFLP method. The SCAR method is used to eliminate the lower reproducibility of RAPD. For the characterisation of selected clones of somatic spruce embryos, cultivated at ourdepartment for a long time, our own clone-specific SCAR markers derived from polymorphous RAPD products were used. The RAPD fragments of DNA were cloned into plasmid vectors, sequenced and specific primers were proposed. Using PCR with these primers we are able to identify effectively the individual clones in our collection.
