The cloning and use of pGREEN based vecttors for the transformation of Pea (Pisum sativum L.)
Original language description
Transformation is emerging as an important crop improvement tool and the engineering of crops with specific traits can be achieved. Among others the transformation of plants with cDNA of viral genes has been shown to result in resistance of the plants tothe corresponding virus. The pGreen series of binary Ti vectors are configured for easy manipulation and to meet the demands of a wide range of transformation procedures for many plant species. Further we optimised the transformation method of pea and the best results were obtained by the sonication-assisted Agrobacterium-mediated transformation of pea embryonic segments. After finishing of the screening of pea virus diseases in field conditions in the Czech republic the appropriate virus gene cDNAs will be cloned into the tested vectors and used for pea transformation.
