Towards long-term 3D culture of leukemic B cells

Chronic lymphocytic leukemia (CLL), the most common adult leukemia in Western countries, is a hematological malignancy of mature B cells. One of the approaches to study CLL is to culture patients' B cells (B-CLL cells) in vitro. However, the primary B-CLL cells are microenvironment-dependent and when cultured in vitro, they shortly un-dergo spontaneous apoptosis. Thus, we aim to develop a 3D system which prolongs the viability of B-CLL cells in vitro. We tested two synthetic materials: hydrogels made of poly(2-hydroxyl methacrylate-co-2-aminoethyl methacrylate) modified with the cell adhesion peptide RGDS [p(HEMA-co-AEMA)-RGDS] with needle-shaped pores; and 0,5% collagen scaffolds. Both materials are transparent and therefore suitable for the inspection with confocal microscopy. Until now, we have tested 3D in vitro culture of HS-5 and MEC-1 cell lines. We studied the optimal cell seeding method, cell adhesion, viability and proliferation. We achieved the most efficient and even cell distribution when using rotation seeding. Importantly, both scaffolds have shown to promote cell adhesion and proliferation of HS-5 as well as MEC-1 cells. The results provide the evidence that the tested materials are suitable for culture of adherent cells as well as cells growing in suspension. This is an important prerequisite for creating the 3D model of CLL, which could possibly serve as a drug-testing platform corresponding to the reality much more than the 2D system.
