In this study we used scanning mutagenesis of the residues within the putative inner pore S6 region of TRPA1 channel with the aim of understanding the process of TRPA1 activation. We identify residues proline 949, asparagine 954 and glycine?(X)3-glycinemotif as structural determinants of TRPA1 activation and deactivation and these findings contribute to our better understanding of molecular mechanisms of pain sensation
