Direct detection of small molecules presents a significant challenge for most biosensor technologies. Consequently, indirect detection formats, such as binding inhibition assays, are commonly employed. In this study, we investigate the relationship between assay parameters and the half-maximal inhibitory concentration (IC50), both theoretically and experimentally, using surface plasmon resonance (SPR) biosensor technology. Our results show that (i) IC50 decreases with decreasing receptor concentration in the sample, asymptotically approaching the equilibrium dissociation constant of the receptor-analyte interaction, and (ii) a similar trend is observed for the limit of detection, suggesting that the proposed theory can be used to estimate the optimal receptor concentration for maximizing biosensor performance
