    Cloning the genes and DNA binding properties of high mobility group B1 (HMGB1) proteins from the human blood flukes Schistosoma mansoni and Schistosoma japonicum

    The parasitic helminthes S. mansoni and S. japonicum contain HMGB1 proteins of primary amino acid sequences and DNA binding properties homologous to vertebrate proteins. Southern hybridization and DNA sequencing revealed a single copy HMGB1 gene. Although DNA binding properties of SmHMGB1 (or SjHMGB1) protein seem to be similar to those previously reported with human HMGB1, we have observed two important differences relative to those observed with the human HMGB1: (i) the inability of the isolated SmHMGB1 domain A to bend DNA (as revealed by T4 ligase-mediated circularization assay), and (ii) higher DNA supercoiling and bending potential of the SmHMGB1 protein as compared to its human counterpart.

    Paraziti S. mansoni a S. japonicum obsahují proteiny HMGB1, které mají podobné aminokyselinové sekvence a DNA vazebné schopnosti jako savčí proteiny. Southern hybridizace prokázala existenci jedné kopie HMGB genu. I když se v mnohém DNA vazebné vlastnosti proteinu SmHMGB1 (nebo SjHMGB1) shodují s lidským proteinem HMGB1, pozorovali jsme tyto odlišnosti: (i) neschopnost izolované domény A proteinu SmHMGB1 ohýbat DNA a (ii) vyšší potenciál proteinu SmHMGB1 superspiralizovat a ohýbat DNA ve srovnání s lidským proteinem.
