DNA and glutathione interactions in cell-free media of asymmetric platinum(II) complexes cis- and trans-[PtCl2(isopropylamine)(1-methylimidazole)]: relations to their different antitumor effects

  • Original language description

The global modification of mammalian and plasmid DNAs by the novel platinum compounds cis-[PtCl2(isopropylamine)(1-methylimidazole)] and trans-[PtCl2(isopropylamine)(1-methylimidazole)] and the reactivity of these compounds with reduced glutathione (GSH)were investigated. The results of this work support the hypothesis that platinum drugs that bind to DNA in a fundamentally different manner from that of conventional cisplatin may have altered pharmacological properties.

Interakce asymetrických komplexů platiny cis- a trans-[PtCl2(isopropylamin)(1-methylimidazol)] s DNA a glutathionem v bezbuněčném prostředí: vztahy k jejich rozdílným protinádorovým účinkům

  • Czech description

Byly studovány globální modifikace savčí a plasmidové DNA novými komplexy platiny cis-[PtCl2(isopropylamin)(1-methylimidazol)] a trans-[PtCl2(isopropylamin)(1-methylimidazol)] a také reaktivita těchto látek s redukovanou formou glutathionu (GSH) v bezbuněčném prostředí. Výsledky této práce podporují hypotézu, že platinové komplexy, které se váží na DNA zásadně rozdílným způsobem oproti konvenční cisplatině, mohou mít pozměněné farmakologické účinky.
