Identification of barley proteins from in-gel digests by MALDI-TOF MS.
Original language description
Proteins were digested from barley leaves, then separated using ID gel electrophoresis and in-gel digested with trypsin. Peptide masses were adjusted through MALDI-TOF and then put into the database searching program. This method is not always successfulbecause the success depends mainly on presence of the protein in the database. In the case of barley only about 20 of genome is known and moreover there exist many variants of barley. In spite of this complications, several proteins were identified. Itproved that the combination of in-gel digestion and MALDI-TOF is suitable for a first attempt of identification of unknown proteins.
