We have descibed for the first time that histone deacetylases (HDAC) inhibitors (trichostatin A, sodium butyrate, MS-275, SAHA, and valproic acid) supress interferon alpha (IFN alpha)-induced up regulation of promyelocytic leukemia protein (PML) at mRNAand protein levels in several human cell lines and in normal diploid skin fibroblasts. Additionaly, HDAC inhibitors blocked an increase of PML nuclear bodies number in response to IFN (alpha)-stimulation. We showed that the effect of HDAC inhibitors on IFN (alpha)-pathway is STAT2- independent.

Jako první jsme popsali, že inhibitory histondeacetyláz (trichostatin A, butyrát sodný, MS-275, SAHA a kyselina valproová) suprimují interferonem alfa (IFN alfa) zprostředkovanou indukci promyelocytárního leukemia proteinu (PML) na úrovni mRNA a proteinuv několika lidských buněčných liniích a v normálních diploidních kožních fibroblastech. Dále inhibitory histondeacetyláz blokovaly nárust počtu PML jaderných tělísek po stimulaci IFN alfa. Prokázali jsme, že efekt histondeacetylázových inhibitorů na IFNalfa dráhu je nezavislý na STAT2.
