Original language description
Retroviral vectors are usually transcriptionally inactivated: this is accompanied by methylation of integrated DNA. CpG islands, common parts of constitutively expressed genes, inhibit methylation and transcriptional suppression of flanking DNA sequences. By inserting a CpG island in front of a heterologous sequence of a reporter provirus containing Rous sarcoma regulation sequences (LTRs) we have constructed a vector resistant to methylation in mammalian cells. Our work represents a new concept of long-term expression maintenance of retroviral vectors for gene transfer.
