Release from tonic inhibition of T cell activation through transient displacement of C-terminal Src kinase (Csk) from lipid rafts.

  • Original language description

Upon triggering of the TCR, PAG is dephosphorylated resulting in loss of Csk from lipid rafts. Tyr-phosphorylation of PAG resumes after 3-5 min, and Csk reassociates with rafts. Cells expressing a mutant Csk without catalytic domain have elevated levelsof TCR-zeta phosphorylation and spontaneous activation of an NFAT-AP1 reporter and stronger responses to TCR triggering. Transient release from Csk-mediated inhibition by displacement of Csk from lipid rafts may be important for normal T cell activation.
