    Differential regulation of the inhibitor of the apoptosis ch-IAP1 by v-rel and proto-oncogene c-rel

    The main aim of this work was to elucidate the mechanism of differential regulation of avian inhibitor of apoptosis (ch-IAP1) by v-rel oncogene and c-rel proto-oncogene. The analysis of ch-IAP1 promoter region revealed four Rel/NF-kappaB binding sites, which are differentially utilized by v-Rel and c-Rel. Although c-Rel was a more potent activator of ch-IAP1 promoter than v-Rel in transient reporter assays, cells overexpressing c-Rel failed to maintain high levels of ch-IAP1 expression. The reduction ofch-IAP1 expression in these cells correlated with the efficient regulation of c-Rel by inhibitory protein IkappaBalpha. The ability of v-Rel to escape IkappaBalpha regulation allows for the gradual and sustained elevation of ch-IAP1 expression directlycontributing to the transforming properties of v-Rel

    Hlavním cílem této práce bylo objasnění mechanismu rozdílné regulace ptačího inhibitoru apoptózy (ch-IAP1) onkogenem v-rel a protoonkogenem c-rel. Analýza promotorové oblasti ch-IAP1 odhalila čtyři vazebná místa pro Rel/NF-kapaB, která jsou v-Rel a c-Relvyužívána rozdílně. I když je v přechodných reportérových testech c-Rel silnějším aktivátorem promotoru ch-IAP1 než v-Rel, buňky se zvýšenou expresí c-Rel nedokázaly udržet vysokou hladinu exprese ch-IAP1. Snížení exprese ch-IAP1 v těchto buňkách bylo vsouladu s účinnou regulací c-Rel inhibičním proteinem IkapaBalfa. Schopnost v-Rel uniknout regulaci IkapaBalfa umožňuje postupné a stálé zvyšování exprese ch-IAP1, což přímo přispívá k transformačním schopnostem v-Rel
