    GATA-1 and c-myb crosstalk during red blood cell differentiation through GATA-1 binding sites in the c-myb promoter

    GATA-1 and c-Myb transcription factors represent key regulators of erythroid development. GATA-1 is upregulated and c-myb protooncogene expression is downregulated when red cell progenitors differentiate into erythrocytes. Here we have employed a culturesystem, that recapitulates erythroid differentiation in vitro, to follow the kinetics of GATA-1 and c-myb expression. We show that c-myb expression is high in progenitors and effectively downregulated at the time when nuclear GATA-1 accumulates and cells differentiate into erythrocytes. Additionally, we identified two GATA-1 binding sites within the c-myb promoter and demonstrate that GATA-1 protein binds to these sites in vitro. Furthermore, GATA-1 represses c-myb expression through one of the GATA-1binding site in transient transfection experiments and this requires FOG-1. Thus, our study provides evidence for a direct molecular link between GATA-1 activity and c-myb proto-oncogene expression during erythroid differentiation

    Transkripční faktory GATA-1 a c-Myb jsou klíčovými regulátory erytroidního vývoje. Když progenitory červených krvinek diferencují v erytrocyty, exprese GATA-1 se zvyšuje a exprese protoonkogenu c-myb se snižuje. Zde jsme využili kultivační systém, ve kterém probíhá erytroidní diferenciace in vitro, abychom sledovali kinetiku exprese GATA-1 a c-myb. Ukazujeme, že exprese c-myb v progenitorech je vysoká a účinně se snižuje v době, kdy se hromadí jaderný GATA-1 a buňky diferencují v erytrocyty. Navíc jsmev promotoru c-myb identifikovali dvě vazebná místa pro GATA-1 a prokázali jsme, že protein GATA-1 se na tato místa váže in vitro. Kromě toho v experimentech s přechodnou transfekcí GATA-1 potlačuje expresi c-myb prostřednictvím jednoho z vazebných míst pro GATA-1 a je k tomu třeba FOG-1. Naše studie tedy prokazuje přímé molekulární spojení mezi aktivitou GATA-1 a expresí protoonkogenu c-myb během erytroidní diferenciace
