The integrity of sperm acrosome is required for normal sperm-egg interaction. Antibodies to acrosomal proteins are still frequently utilized to check the acrosomal state. We studied acrosomal proteins in boar and human spermatozoa by using of Hs-8 and Hs-14 monoclonal antibodies (MoAbs) which were prepared in our laboratory.The intra-acrosomal proteins recognized by Hs-8 and Hs-14 MoAbs are stable upon capacitation and even after induction of acrosome reaction. The role of pHs-8 and pHs-14 proteins in sperm-egg binding was investigated by in vitro binding assay of boar spermatozoa and porcine oocytes. The Hs-14 relevant protein does not participate in gamete binding while the Hs-8 detected protein is a secondary binding protein

Celistvost akrozomu savčích spermií hodnocená monoklonálními protilátkami proti vnitřním proteinům akrozomu a úloha těchto proteinů ve vazbě spermie a vajíčka

Celistvost akrozomu spermie je nezbytná pro normální interakci spermie s vajíčkem. K ověření stavu akrozomu se ještě stále často používají protilátky proti proteinům akrozomu. Studovali jsme akrozomální proteiny pomocí protilátek Hs-8 a Hs-14 připravených v naší laboratoři. Proteiny uvnitř akrozomu rozpoznané Hs-8 a Hs-14 jsou při kapacitaci a dokonce i po indukci akrozomové reakce stabilní. Byla zkoumána úloha proteinů pHs-8 a pHs-14 ve vazbě spermie na vajíčko pomocí vazebného testu kančích spermií aprasečích oocytů in vitro. Protein zjištěny Hs-14 se vazby gamet neúčastní, zatímco protein detekovaný Hs-8 je proteinem sekundární vazby
