    Dysregulation of Src family kinases in mast cells from epilepsy-resistant ASK versus epilepsy-prone EL mice

    ASK mouse strain mast cells proliferated more vigorously than EL strain cells in response to IL-3 and stem cell factor. Although ASK mast cells degranulated less vigorously than EL mast cells upon stimulation with IgE and Ag, ASK cells produced and secreted several-fold more TNF-alpha and IL-2 than EL cells. Consistent with the similarities of these ASK and EL mast cell responses with phenotypes of lyn(-/-) and wild-type mast cells, respectively, Lyn activity was reduced in ASK cells. In addition to theimpaired Lyn activity, ASK cells just like lyn(-/-) cells exhibited reduced Syk activity, prolonged activation of ERK and JNK, and enhanced activation of Akt. Furthermore, transmembrane adaptor protein PAG that associates with Lyn was hypophosphorylatedin ASK cells. Fyn was hyperactivated in ASK cells. Therefore, Lyn-dependent phosphorylation of PAG negatively regulates Src family kinases.

    Mastocyty myšího kmene ASK proliferují v závislosti na IL-3 a SCF silněji než buňky kmene EL. Ačkoli mastocyty ASK degranulují po stimulaci IgE a antigenem méně, sekretují několikrát více TNF-alpha a IL-2 než buňky EL. V souladu s podobností těchto ASK and EL mastocytů s fenotypy lyn(-/-) vs. wt masocyty, byla pozorována snížená aktivita Lyn u buněk ASK. Navíc ASK buňky právě jako lyn(-/-) buňky měly sníženou aktivitu Syk, prolongovanou aktivitu ERK a JNK, a zvýšenou aktivaci Akt. Adaptorový protein PAGasociující s Lyn byl hyperfosforylován v buňkách ASK. Kinasa Fyn byla v těchto buňkách také hyperaktivovaná. Lyn-dependentní fosforylace PAG tedy negativně reguluje kinasy skupiny Src.
