    ASLV family retroviruses contain 5 highly related envelope subgroups (A-E). Susceptibility or resistance of cells to infection by these ASLVs is given by 3 loci in chicken. The tvb gene encodes subgroup B, D, E ASLV receptors. Wild-type TvbS1 receptor confers susceptibility to subgroup B, D, E ASLVs. In this study the genetic defect underlying altered susceptibility of inbred chicken line M to infection by ASLV(B), (D), (E), C125S substitution in tvb-encoded TvbS1 receptor in this line, was identified.We show that this substitution significantly reduces susceptibility of line M cells to infection by ASLV(B) and (D) and virtually eliminates susceptibility to ASLV(E) infection in cultured cells and the incidence and growth of ASV-induced sarcomas in chickens. It also significantly reduces binding affinity of TvbS1 receptor for subgroup B, D, E ASLV envelope glycoproteins. These results are first demonstration of possible role of the Cys-rich domain 3 in the function of Tvb receptors.

    Jednoaminokyselinová substituce v receptoru TvbS1 snižuje susceptibilitu k infekci ptačími sarkomovými a leukosovými viry podskupin B a D a podmiňuje resistenci k podskupině E in vitro a in vivo

    Ptačí sarkomové a leukosové viry (ASLV) tvoří pět interferenčních podskupin (A až E) a využívají pro vstup do buněk tří různých receptorů. Viry podskupin B, D a E vstupují do susceptibilních buněk přes receptor Tvb. Identifikovali jsme mutaci v receptoruTvb, která podmiňuje semiresistentní fenotyp drůbeží inbrední linie M vůči virům podskupiny B a D a téměř úplnou resistenci ke skupině E. Mutace spočívá v záměně cysteinu v posici 125 v cysteinem bohaté doméně 3, která byla doposud pokládána za nedůležitou pro vazbu virových obalových glykoproteinů. Předpokládáme, že při absenci cytosinu 125 dochází k prodloužení receptorové molekuly a navázaný retrovirus je tak příliš daleko od membrány než aby mohl účinně fúsovat s cílovou buňkou.
