Original language description
A new method for selection of aptamers specific for Taq DNA polymerase was developed. The method consists of several steps. In the first step aptamers, which bind nonspecifically to bovine serum albumin are removed from aptamer library. Next, aptamers binding to Taq polymerase are amplified directly in polymerase chain reaction (PCR) tubes and tested in a binding assay. Aptamers which bind to Taq DNA polymerase are then amplified in symmetrical PCR and cloned in plasmid vector. After amplification of the vector in bacteria the aptamer inserts are sequenced. Based on the sequence the aptamers are prepared synthetically and tested.
