    Viscoelastic properties of collagen matter assessed by slit rheometer and pressure hole method

    Collagen is the most abundant protein in human body and has got a large usage not only in the medicine or tissue engineering, but also in the food industry. The rheological parameters of the collagen water solution with high mass content of the collagen (up to 9 %) were evaluated. Determination of rheological properties is key knowledge in the process of designing the production line processing the collagen material. The tested material resembles with its structure a bread dough and is solid-like when no stress is applied. Its unique structure caused by high dry matter content brings up challenges in the determination of rheological properties. The own-constructed extrusion rheometer which is suitable to operate at high shear rates was used for experiments. The experiments can be done at large shear rates range (γ ̇= 10^2 -10^4 s^(-1)) which is gained by variable speed of hydraulically driven piston which is pushing the measured material through capillary. In addition, the construction of the rheometer allows to change the geometry of the capillary head. Rectangular capillaries with variable heights (H=1, 2, 4, 8 mm), convergent-divergent capillary or capillary for hole-pressure measurements are used. The rheometer is equipped with 5 pressure transducers along the capillary head and the position of the piston is monitored as well. The rheological parameters are evaluated by modified Herschel-Bulkley model. Although the collagen dough itself is considered incompressible, effects of compressibility can be observed during experiments (e. g. the outflow of material from the capillary when the piston is no longer moving). This effect is caused by the present air bubbles, the theory of two-phase flow is presented as well. The methodology for evaluation of viscoelasticity is examined by measurements of exit pressure and hole pressure method. The determination of the dynamical yield stress from relaxation of pressures after stopping of the piston movement. This independent evaluation. Although the dynamical yield stress is evaluated from the same experiment as coefficient of consistency K and the flow index n, the evaluation is absolutely independent since it is executed from different point of the experiment.
