Chitinolytic bacterial populations in gastrointestinal tract of wild ruminants
Original language description
In this study we focused on bacteria from the animal digestive tract with an ability to utilize chitin. As cultivation media we used M-10 medium with colloid chitin as source of energy. Cultivation was carried out under anaerobic conditions. Chitinolyticbacteria were isolated from enrichment culture with faeces of different wild ruminants (fallow deer, Pere David's deer, Roth schild's giraffe). Isolates were selected accoring to their activity towards CM chitin and then characterized. The identification of strains with chitinolytic activities was made due to sequencing method. For further characterization of biochemical ability to degrade chitin substrate we will perform polyacrylamide gel electrophoresis of proteins, which are produced by chitosan degradation. The visualization of the chitinolytic enzymes will be achieved by zymography technique.
