The methods of biomedical research in mapping genes that modify immunological parametres and in analysis of gene expression
Original language description
Biomedical methods and technology are the fundamental part of biological researches. In the present work we aimed to apply combination of different methods to investigate genetic control of leishmaniasis. Leishmaniasis is a diseases caused by Leishmania parasite. In our lab we use recombinant congenic strain of mice to dissect genes controlling pathology in Leishmania infection. We have analyzed 209 F2 hybrids between strains OcB-43 and O20 and genotyped using 7 markers by PCR on four chromosomes. In order to identify genes controlling response to the diseases we also tested expression of selected genes using Real-time PCR. Data were analyzed using Statistica software. We identified 3 loci controlling size of secondary lymphoid organs. The results of expression analysis showed that expression of selected genes depend on genetic background of the host
