In our study we aimed to find the universal procedure which would enable us to get DGGE profiles from larger set of samples where the soils differed substantially in contents of organic matter. Fraction of humic substances displays similar properties toDNA hence it can be uneasy to remove them during DNA isolation process and the substances consequently inhibit PCR. Additional cleaning of isolated DNA appears to be necessary. for most of our samples, two cleanings of isolated DNA (Wizard DNA clean-up systém, Promega) and subsequent tenfold dilution were sufficient. The most problematic sample needed three cleaning steps and tenfold dilution. Our experiences imply that dilution is more useful than additional cleaning for some samples.

Byl ověřován postup isolace bakteriální DNA z velmi humosních stepních půd. Huminové látky ztěžovaly isolaci DNA z půd i inhibovaly následnou proceduru PCR. Práce prokázala, že dodatečné přečištění získané DNA je nezbytné. Z některých typů vzorků je všaknemožné získat požadovanou DNA universálním postupem.
