    Temperature gradient gel electrophoresis (TGGE) in study of environmental samples: first experiences

    TGGE is an efficient method which allows us to study both composition and dynamics of complex microbial communities without the necessity of cultivation. Sample´s total DNA isolation and consequent PCR amplification of desired gene´s fragment need to becarried out before the TGGE experiment. Such pretreatment provides us with the population of gene fragments of the identical length from virtually all the strains present in the sample, and differing in their exact sequences. The DNA sequence variation in obtained gene fragments is a base for subsequent TGGE separation and identification of particular bacterial strains. Currently an equivalent method called denaturing gradient gel electrophoresis (DGGE) is used more often usually performed on D-code instrument (Bio-rad). In our laboratory we try to adopt TGGE instrumented on TGGE-Maxi system (Biometra) where the procedure is different particularly in the gel preparation and some other parameters.
