    Infections in chronic wounds can alter acidic pH of healthy skin environment to alkaline pH. Rifampicin (RIF) is the only potent antibiotic against most infections, but its poor solubility, low bioavailability, pH and temperature sensitivity significantly limit its therapeutic e↵ect. To circumvent these obstacles, two innovative biodegradable polyelectrolyte nanocarriers (APENs and WPENs) composed of amphiphilic chitosan core, ionic cross linking dextran sulfate and polyethyleneimine shell were prepared using layer-by-layer self-assembly of polyelectrolytes at alkaline pH. Thereafter, physicochemical characteristics of the modified polymers were evaluated by FT-IR and elemental analysis. CQA’s including PENs’ morphology, size, z-potential, drug loading, release profile and degradation were evaluated by SEM, TEM, DLS and UV-Vis spectrophotometry, respectively. The results confirmed that pH-insensitive cross-linker caused controlled release of RIF, and strong hydrophobic interaction among RIF and hydrophobic moieties of modified CS led to significantly increased z-potential, compact structures and low degradation of RIF against alkaline pH.
