    The main objective of this paper is to reveal and to enhance our understanding of how entrepreneurial universities, as higher education institutions (HEIs), significantly contribute to fostering knowledge-based economic growth and supporting the sustainability of social innovation ecosystems. It also seeks to illustrate how the entrepreneurial universities&apos; prominent processes and strategies enhance Life Cycle Sustainability Assessment (LCSA), encourage regional knowledge spillovers, and support social innovation ecosystems, ultimately facilitating better knowledge transfer between industry and academia through their third mission initiatives. The existing knowledge spillover within the regions has inspired the creation of new knowledge, exchange of knowledge across-disciplinary areas of research that empowers regional small firms, i.e., industry and academia, i.e. higher education institutes (HEIs) to face the economic, social, and industrial turbulence.Till now, higher education institutions (HEIs) have been evolving into full-fledged managerial entities that create an economic impact on local, regional, and national scales. Currently, entrepreneurial universities&apos; emerging perspectives received considerable attention due to their key enabling inspirations that enable HEIs and SMEs in knowledge production, inclusive growth, and sustainable development. Moreover, it facilitates the regional innovation system to promote the tacit use of knowledge in the regions, retention in SMEs, and the dynamics of knowledge.Finally, this research emphasized the important advantages that entrepreneurial universities bring to the social innovation process, which in turn shapes the perception of enhancing regional knowledge development. Entrepreneurial universities impact various concepts, including life cycle sustainability assessment, regional knowledge spillover, and social innovation ecosystems, to improve knowledge transfer, develop spin-off companies, and connect new knowledge with its practical applications in industry and society. Moreover, the study illustrates how the entrepreneurial discovery processes (EDP) of entrepreneurial universities play a vital role as a means of collaborative knowledge exchange between industry and academia in the knowledge economy. They act as an essential enabler, influencing the conditions that promote sustainable entrepreneurship and the spread of knowledge in a particular area.
