Ancient DNA study of Bronze and Iron Age human populations from central Europe
Project goals
Our proposed project focuses on research of human populations from the Bronze and Iron Age, from the region of Czech Republic and Poland, and it covers the period of last two millennia before current era. The principal aim of our study is to describe the genetic structure and variability of prehistoric populations together with the inter-population relations. This will allow us to (a) confirm or refuse the hypothesis of population continuity in central Europe during the studied period, (b) reconstruct the possible migration routes of the prehistoric populations. To reach these goals we will analyze the nuclear genome and mitochondrial DNA of the samples. The specific objectives on population level include sequencing of full nuclear genomes, followed by analyzing the principal components of genetic variability and population admixture analysis with the use of SNP (single nucleotide polymorphism) markers. Our objectives for sample level include typing the genetic sex of the samples, establishing the mitochondrial haplogroup, and kinship analysis for the samples. The research part of our project is already supported by grants of Polish national grant agency (National Science Center) and by ELIXIR CZ consortium. Secondary aims of this project consist of establishing more efficient cooperation between the partner institutions, including the transfer of knowledge and experience, supporting professional growth of young researchers, and searching for the opportunities of broadening our research collaboration in the future.

Výzkum starodávné DNA lidských populací z doby bronzové a železné z oblasti střední Evropy

Náš projekt se zabývá výzkumem lidských populací z doby bronzové a železné na území České republiky a Polska, přibližně v období posledních dva tisíce let před naším letopočtem. Hlavním cílem je popsat genetickou strukturu a variabilitu zkoumaných populací a zároveň mezipopulační rozdíly. To nám umožní (a) potvrdit nebo vyvrátit hypotézu o populační kontinuitě ve střední Evropě během studovaného období, (b) zrekonstruovat možné migrační dráhy prehistorických populací. Tyto cíle budou dosaženy díky analýze jaderného genomu a mitochondriální DNA z prehistorických vzorků. Detailní cíle na populační hladině zahrnují tedy sekvenaci jaderných genomů a následnou analýzu hlavních komponent populační variability a analýzu mísení populací (admixture) za využití informací z SNP (single nucleotide polymorphism) lokusů. Na hladině jednotlivých vzorků nás pak bude zajímat jejich genetické pohlaví, variabilita mitochondriálních haploskupin a příbuznost mezi vzorky pocházející z jedné lokality (kinship). Výzkumná část projektu je již podpořená projekty polské grantové agentury (National Science Center) a konsorcia ELIXIR CZ. Další cíle toho grantového projektu z hlediska navázání užší spolupráce mezi partnerskými institucemi zahrnují transfer znalostí a zkušeností, rozvoj mladých výzkumníků zapojených do projektu, a hledání možností rozšíření naší vědecké spolupráce v budoucnu.
