Signalling during Pollen-Pistil interaction preceding fertilization; the role of short- and long-distance secreted signals of fertilization

Project goals

The delivery of non-motile male sperm cells for fertilization critically depends on precise signaling between the growing pollen tube and the female gametophyte conveyed by secreted ligand molecules. We have reconstructed tobacco pollen tube growth through the pistil (semi-in vivo, SIV) and captured over 800 protein groups representing secreted ligands and receptor of the pollen tubes. Their analysis provided evidence that pollen tube growth through the pistil endures diverse signaling pathways including small GTPase-mediated signal transduction, membrane fused secretion and cell wall matrix remodeling. Proteins belong to plant defensin-like family (DEFL), LORELEI-like GPI-anchored protein 3 (LLG3), Thionin-like protein, plant lipid transfer proteins (LTPs) pollen Ole e allergen family, small GTPases as well pectinases, were among the identified pollen tube secreted proteins. Their secretion was largely perturbed in the yip4a;4b double mutant. Our project proposal aim at dissecting the signaling role undertaken by identified secreted proteins during pollen tube-ovule targeting.

    Signalizace mezi pylem a pestíkem předcházející oplození; úloha sekretovaných signálů působících na kratší a delší vzdálenost

    Doručení nemobilních samičích spermatických buněk k vajíčku při oplození je kriticky závislé na přesné signalizaci mezi rostoucí pylovou láčkou a samičím gametofytem, sprostředkovanou sekretovanými ligandmi. Semi-in vivo metodou jsme napodobnili růst láčky pestíkem a následně jsme identifikovali přes 800 proteinových skupin, které reprezentují sekretované ligandy a receptorové pylové láčky. Jejich analýza nám poskytla důkazy, že růst láčky pestíkem si vyžaduje speciální signalizační dráhy, mezi které patří signalizace pomocí malých GTP-áz, sekrece po fúzi membrán, proteiny zapojené remodelace matrix buněčné stěny. Identifikované sekretované proteiny z pylových láček patří do rodin defensin-like (DEFL), LORELEI-like GPI-anchored proteiny 3 (LLG3), thionin-like proteiny, rostlinné lipid transferové proteiny (LTP), rodina pylových Ole e alergenů, malých GTPáz a pektináz. Jejich sekrece byla výrazně odlišná v dvojitém mutantovi yip4a;4b. Návrh našeho projektu má za cíl popsat signální roli sekretovaných peptidů počas “zacílení” vajíčka pylovou láčkou.

    The project has enlarged the knowledge of the role of small proteins secreted by pollen tube, mechanisms of ligand-receptor interactions and has also resulted in an identification of proteins secreted by ovule. The aims were fulfilled and some results were obtained in addition to plan. Five papers in prestigeous journals were published. The funds were used in planned structure and amounts.
