    Adhese, růst a diferenciace kostních a cévních buněk na uhlíkových allotropech

    Cílem tohoto mezioborového projektu je připravit bioaktivní uhlíkové vrstvy perspektivní pro povrchové modifikace biomateriálů užívaných v tkáňovém inženýrství, zejména pro kostní implantáty. Tyto vrstvy vzniknou jednak naprášením či napařením z uhlíkového terče, jednak nanesením allotropů uhlíku, jako jsou fullereny, nanotrubičky či nanodiamanty. V podmínkách in vitro budou osazeny osteoblasty a cévními hladkými svalovými buňkami. Očekáváme, že vrstvy svou nanostrukturou podpoří adsorpci vitronektinu,tj. proteinu extracelulární matrix zprostředkujícího preferenční adhesi osteoblastů, která bude dále posílena i funkcionalizací vrstev kyslíkatými skupinami či aminoskupinami. Vrstvy složené z fullerenů a nanotub budou naroubovány i ligandypro adhesní receptory buněk (např. oligopeptidy s aminokyselinovou sekvencí RGD či oligopeptid KRSR, specifický pro osteoblasty). Adhese, růst a diferenciace buněk budou regulovány i pomocí hierarchicky organizované mikro- a nanostruktury povrchů,
