Structual antigens of papovaviruses: fuctions , cellular interactions and utilisation for gene therapy purposes

Project goals

The aim of the presented project is to develop a carrier system for DNA transfer into mammalian cells (and organisms) for gene therapy purposes, utilising artificially made papovavirus empty capsid-like particlaes. Th egreat advantage of these DNA transfer vehicles would be that capsid-like particles produced by self-assembly from either one, two or three structural proteins, massively produced from recombinant baculoviruses in insect cells, can be easily purified into homogenity and provide as with well defined system, without unwanted viral sequences. In our preliminary experiments we have shown that artificial capsid-like particles are able to encapsidate 'foreign' DNA and delivery it for functional expression into nuclei of rat and human cells, Theproposals involve also basic research of functions of individual polyoma- and papilloma- virus late antigens and their interactions with host cell structures in viral life cycle.

    Strukturní antigeny papovavirů: Funkce, buněčné interakce a využití pro účely genové therapie

    Cílem předkládaného projektu je vyvinout systém pro přenos DNA sekvencí savčích buněk pro nápravu genetických poruch využívající arteficiálně připravené kapsidové struktury papovavirů. Tento systém nabízí řadu výhod. Struktury prázdných kapsid mohou býtsnadno vyčištěny do homogenity ze strukturních proteinů produkovaných v hmyzích buňkách bez jakéhokoli kontaktu s papovavirovými genomy. Bylo ukázáno, že takto připravené partikule jsou schopny enkapsidovat in vitro cizí DNA a přenést ji do myších i lidských buněk. Projekt zahrnuje též základní výzkum funkcí jednotlivých strukturních antigenů v životním cyklu polyoma- a papilloma- virů a jejich interakcí s buněčnými strukturami.
