Cytokinin metabolism in tobacco chloroplasts and its regulation

Project goals

The development and function of the plant-specific organelle - chloroplast - is predominantly affected by plant hormones cytokinins (CKs). To obtain the relevant picture of CK metabolism and its regulation in chloroplasts, the isolated organelle isnecessary to be used. This project is focused on the study of the occurrence and metabolism of CKs in chloroplasts isolated from tobacco leaves. To evaluate the regulatory mechanisms involved in CK metabolism, the levels of CKs will be modified (1) byexpression of specific genes [gene coding for isopentenyl transferase (CK biosynthetic enzyme) under the control of light-inducible ssU (Rubisco) and tetracycline-inducible promoters and gene coding for ß-glucosidase (enzyme which releases active CKsfrom storageforms - O-glucosides)) under the control of constitutive CAMV 35S promoter] and (2) by application of the inhibitors of CK metabolic pathways - N-glucosylation (papaverine and roscovitine) as well as of cytoplasmic isoprenoid (lovastatin) or

    Metabolismus cytokininů a jeho regulace v chloroplastech tabáku

    Vývoj a funkce chloroplastů je rozhodujícím způsobem ovlivňována rostlinnými hormony cytokininy (CK). Tento projekt je zaměřen na sledování metabolismu CK a jeho regulace v chloroplastech isolovaných z listů tabáku. Ke studiu mechanismů regulacemetabolismu CK bude využito ovlivnění hladiny CK: (1) expresí genů kódujících enzymy, které se účastní přeměny CK [genu pro isopentenyltransferasu (ipt), jehož produkt zvyšuje hladinu CK, a genu pro ß-glukosidasu (Zmp-60.1), jehož produkt uvolňujeaktivní CK ze zásobních O-glukosidů]. Gen ipt bude použit pod kontrolou inducibilních promotorů (aktivace světlem nebo dereprese tetracyklinem), gen Zmp-60.1 bude pod kontrolou konstitutivního promotoru 35S. (2) aplikací inhibitorů metabolických drah CK,jak N-glukosylace (papaverinu a roscovitinu), tak tvorby isoprenoidů (cytoplasmatická dráha - lovastatin a plastidová dráha - fosmidomycin).
