The role of cytokinin/abscisic acid balance in abiotic stress response and recovery
Project goals
The role of plant hormones cytokinins (CK) during the abiotic stress (drought, heat shock and their combination) response and recovery will be evaluated by comparison of photosynthetic activity, pigment composition and water loss in tobacco plants with elevated and suppressed levels of endogenous CKs. Metabolism of CKs and "stress hormones" abscisic acid and ethylene will be monitored in plants over-expressing CK biosynthetic gene (SHO - shooting) and gene for CK deactivation enzyme (CKX3 - cytokinin oxidase/dehydrogenase), respectively. CK and ABA concentrations will be determined also in chloroplasts, a site of their synthesis. Chloroplast and mitochondria ultrastructure will be estimated microscopically. Three typical stress responses - expressionofselected stress-inducible genes (coding heat shock proteins), stimulation of antioxidant defense system (expression level and activity of selected antioxidant enzymes) and polyamine formation will be monitored. Úloha rovnováhy mezi cytokininy a kyselinou abscisovou při abiotickém stresu a během následného zotavení

Úloha rostlinných hormonů cytokininů (CK) při abiotickém stresu (sucho, zvýšená teplota, jejich kombinace) a během následného zotavování bude studována v rostlinách tabáku porovnáním chování rostlin se zvýšenou hladinou CK (zvýšená exprese biosyntetického enzymu CK: SHO) a sníženou hladinou CK (zvýšená exprese genu kódujícího hlavní enzym pro deaktivaci CK: cytokinin oxidázu/dehydrogenázu CKX3). Bude sledována hladina CK, kyseliny abscisové (ABA) a etylénu. Hladina CK a ABA bude stanovena i v chloroplastech, které jsou jedním z míst jejich biosyntézy. Vliv stresu na strukturu chloroplastů a mitochondrií bude sledován elektronovou mikroskopií. Bude studován vliv CK na tři procesy, které jsou stresem silně ovlivněny: exprese genů indukovatelných stresem(vybrané geny kódující "heat shock" proteiny), stimulace antioxidativních enzymů (jak na úrovni exprese, tak aktivity) a tvorba polyaminů. Získané výsledky přispějí k pochopení mechanismů odezvy rostlin na abiotický stres.
