    Vliv exprese genu pro zeatin O-glukosyltransferázu z fazolu na toleranci vůči vodnímu stresu u rostlin tabáku

    Cílem tohoto projektu je ověřit možnost zvýšení tolerance rostlin vůči vodnímu stresu ovlivněním metabolismu rostlinných hormonů cytokininů (CK). Celková hladina CK bude v rostlinách tabáku navýšena expresí genu pro zeatin O-glukosyltransferasu (ZOG) izolovaného z fazolu. Transformované rostliny obsahují značné množství CK v neaktivní, ale snadno mobilizovatelné formě O-glukosidů. Koncentrace fyziologicky aktivních basí, ani morfologie není v těchto transformantech výrazně ovlivněna. Chování rostlin bude sledováno při mírném, středním a silném vodním stresu a během následné rehydratace. Úroveň stresu bude stanovena pomocí parametrů vodního režimu, fotosyntézy a hladiny kyseliny abscisové.Vliv exprese genu pro ZOG z fazolu pod konstitutivním promotorem35 S v rostlinách tabáku bude sledován na úrovni RNA (RT-PCR, Northern analysy), proteinů (Western analysy, dvojrozměrná elektroforesa), aktivity enzymu a hladiny jednotlivých CK Rovněž bude stanovována aktivita hlavního degradačního enzymu CK
