    High complexity and dual genetic origin of mitoribosomes, ribonucleoprotein complexes responsible for proteins synthesis in mitochondria, are reflected in their intricate assembly processes relying on multiple assembly factors. Early stages of mitoribosome biogenesis, unlike late steps, remain obscured. Here, we will utilize the protist Trypanosoma brucei to understand the early assembly of the small mitoribosomal subunit (mtSSU), which is difficult to study in other model organisms. We will combine biochemical and proteomic approaches to identify and characterize early assembly factors widespread in eukaryotes and specific to trypanosomatids. Using cryoEM, we will determine structures of early assembly intermediates to reveal the conserved and group-specific mechanisms of early biogenesis. We will also rationalize a unique direct link we found between an mtSSU precursor and U-indel RNA editing. Our results will provide grounds for development of diagnostic or therapeutic tools against mitoribosome-associated mitochondrial disorders and diseases caused by trypanosomatids parasites.
