    Pulmonary Hypertension: A Multitarget Approach Using Tailored Galectin Inhibitors

    Galectins are soluble human lectins with many (patho)physiological effects. Galectin-1, -3, -8 and -9 have been implicated in the pathogenesis of cardiovascular diseases such as pulmonary hypertension (PH). The common manifestation of the different subtypes of this heterogeneous disease is pulmonary vascular remodeling. The pathways of galectin action in the development and progression of PH are poorly defined. We intend to evaluate the therapeutic potential and pharmacokinetics of newly prepared tailored glycopolymer galectin inhibitors with proven inhibitory potential to galectins in a series of in vitro and in vivo assays. In vitro studies with pulmonary artery endothelial cells, smooth muscle cells, adventitial and cardiac fibroblasts will complement in vivo assays with rat models of hypoxia- and monocrotaline-induced PH. We aim to elucidate the mechanisms of PH pathophysiology related to individual galectins and to assess the potential of their pharmacological inhibition as a novel therapeutic strategy for PH.
