    Library of engineered glycosidases for the preparation of bioactive glycans

    The aim of this project is to construct a toolbox of genetically modified glycosidases with desired catalytic properties, in particular enhanced synthetic capability, enhanced regioselectivity and/ or substrate specificity, and to use these enzymes in cascade transglycosylation reactions to afford a library of tailored oligosaccharide ligands of biomedically relevant galectins. Due to the broad substrate specificity of engineered glycosidases, the prepared tailored oligosaccharides will bear suitable linkers for conjugation to multivalent biocompatible water-soluble carriers that will afford multivalent glycoconjugates with high affinity (avidity) and possibly also high selectivity for particular galectins. Affinity of prepared glycostructures will be tested by ELISA-type assays and other methods. The resulting glycoconjugates will be prospective for application in the therapy or prevention of diseases linked to galectin overexpression, especially tumor progression

  • Project results evaluation

    A library of new mutant and native enzymes for oligosaccharide synthesis was prepared. Selected carbohydrates were conjugated to multivalent carriers including calixarenes and their affinity to a series of human galectins was determined by ELISA and biolayer interferometry. The results of structure-function studies can be used in further biomedical research.
