The project focuses on developing experimental glycopolymer-based biosensors for detecting galectins, carbohydrate-binding proteins linked to, e.g., pancreatic and brain cancers. These fatal diseases currently lack effective non-invasive diagnostic tools for early detection. We will design glycopolymer brushes derived from hydrophilic poly(N-(2-hydroxypropyl)methacrylamide), functionalized with complex carbohydrate ligands with a high affinity and selectivity to target galectins, especially Gal-1 and Gal-3. Chemo-enzymatic synthesis of carbohydrate ligands and controlled polymerization techniques will help achieve precise glycopolymer architectures. Surface functionalization strategies are employed to optimize binding efficacy and antifouling properties, ensuring accurate detection in complex biological fluids like blood plasma. We will assess the relationship between the composition and characteristics of glycopolymer brushes, their binding efficacy and selectivity to target galectins. Finally, we will assess the applicability of the experimental biosensors for real patient samples.
