Depression is a prevalent psychiatric disorder often exhibiting substantial resistance to treatment. This study aims to examine the impact of chronic activation of parvalbumin (PV) interneurons in the medial prefrontal cortex (mPFC) on depressive-like behavior, nociception, corticosterone levels, and neural activity. Using PV-Cre transgenic mice, we will employ chemogenetic tools to selectively activate PV interneurons, followed by behavioral assessments, histological evaluation, and electrophysiological recordings. Given their potential regulatory role, vasoactive intestinal peptide (VIP) interneurons will be investigated for their capacity to counteract PV hyperactivity and normalize mPFC excitability. Additionally, we will explore ketamine administration and optogenetic stimulation of VIP interneurons as potential therapeutic interventions. This research may inform the development of novel circuit-based treatments for depression.
