Infections resulting from injuries can lead to severe, potentially fatal outcomes, making their prevention a critical challenge in modern healthcare. To prevent infections, wound dressings are often functionalized with antibiotics or antimicrobial agents. However, the overuse of such agents has accelerated the emergence of multidrug-resistant bacteria. Hence, the target of this project proposal is to develop a new type of superstructure PCL nanofibers with unique nanotopography serving as drug carriers of one (NFSK-SDC) or two (NFSK-DDC) antimicrobial agents. These will combine their bactericidal effects based on both mechanical (e.g., nanopillar puncture) and chemical influences, using lower but effective amounts of antimicrobials, thereby preventing drug overuse. This approach is based on a hypothesis that (i) by combining morphological effects, such as fiber diameter, and especially nanotopography, pathogens' ability of adhesion and colonization of the material by microbes may be decreased, and (ii) the required antimicrobial load in the material can be reduced.
