    Although immune checkpoint inhibition (ICI) by PD-1 blockade has revolutionized immunotherapy, it benefits only in a subset of patients. Non-small cell lung carcinoma (NSCLC) is typically ICI-responsive, immunologically hot, and enriched in tertiary lymphoid structures (TLS) and stem-like CD8+ T cells, whereas high-grade serous ovarian carcinoma (HGSOC) is generally the opposite. Based on our preliminary data, we hypothesize that immunosuppressive tumor-associated macrophages (TAMs) inhibit T-cell responses to ICI therapy. To test this, we will investigate the mechanisms of TAM-mediated ICI-resistance in human HGSOC and NSCLC samples, as well as unique murine models of HGSOC using a multidisciplinary approach. In parallel, we will assess whether targeting IFN/STAT1 signaling, inhibiting PTPN1/2 phosphatases, and/or promoting IL-2 signaling can overcome ICI resistance in HGSOC. This project will improve our understanding of immunosuppressive mechanisms in the tumor microenvironment and identify novel strategies to increase the efficacy of cancer immunotherapy.
