Asymmetric Organocatalysis as a Gateway to Inherently Chiral Compounds

  • Annotation in Czech

Inherently chiral compounds, such as macrocyclic polyaromatics, serve as a versatile platform for applications, including enantioselective recognition agents, molecular switches, host-guest systems, drug/gene delivery platforms, molecular devices, and other smart materials. However, the potential of enantiopure inherently chiral compounds remains limited due to the challenges associated with their synthesis. The goal of this project is to advance asymmetric organocatalysis by developing new methodologies for synthesizing enantiomerically enriched inherently chiral molecules. The first part of the project focuses on constructing chiral pillar[5]arenes, [2]rotaxanes, and calix[4]arenes via catalysis with chiral N-heterocyclic carbenes. The second part explores Brønsted acid and Lewis base-catalyzed approaches, including desymmetric reductive amination and asymmetric esterification of calix[4]arenes at distinct rim positions, as well as asymmetric intramolecular cyclization leading to the formation of novel eight-membered saddle-shaped molecules.
