Electrophilic compounds are an integral part of the regulatory system in biological organisms. They are produced endogenously and some have been proposed as drug candidates. These transient reactive molecules are subject to rapid decay and nonspecific interactions, which is challenge for the development of new analytical strategies for their determination, or direct screening, identification/characterization. In the project proposal, we present a novel approach based on the application of chemoselective nucleophilic probes (NuP) suitable for trapping electrophilic compounds in mammalian cells. NuPs were designed to form stable adducts with electrophiles through the thiol group. The detection step is based on the identification of the electrophile adduct with NuPs, bearing fluoride moieties with a unique pattern suitable for mass spectrometric-based analysis. The strategy developed may contribute to the identification of hitherto unknown electrophilic compounds particularly in the context of inflammatory processes and oxidative stress.
