    Epigenetická regulace genové exprese v transgenních a endogenních lokusech vyšších rostlin

    Rostlinné transgeny často oproti teoretickým předpokladům vykazují sníženou expresi v důsledku epigenetických modifikací (například metylace DNA), což negativně ovlivňuje aplikaci geneticky modifikovaných rostlin v zemědělství a biotechnologiích. Asociace mezi metylační modifikací DNA a umlčováním genové exprese byla ověřená na mnoha živočišných i rostlinných modelech. V předkládaném projektu navrhujeme provést hlubší analýzu našeho původního pozorování - šíření metylace DNA v posttranskripčně umlčenémtransgenním lokusu Nicotiana tabacum v průběhu kulivace buněk in vitro. Na modelových transgenních liniích budou zkoumány vývojové, genetické a evoluční aspekty tohoto fenoménu. Bude sledován vliv epigenetických modifikací na expresi reporterového genu pro bakteriální neomycinfosfotransferázu. Linie obsahující epimutované alely transgenu budou dále podrobeny analýze z hlediska schopnosti induktoru umlčovat expresi homologního lokusu in trans. Budou hledány paralely v epigenetickém
