Inhibitors of insect digestive enzymes associated with natural defence of plants against pests

Project goals

Plant inhibitors of insect digestive enzymes play an important role in natural defense of pants. Spectrum of both inhibitors and enzymes is changing during the complex process of plant-insect interaction (wound-induced inhibitors, inhibitor-induced enzymes) The potential of protease and alfa-amylase inhibitors to block the digestive enzymes of insects will be studied both in vivo and in vitro. The project includes isolation and characterization of selected plant inhibitors from potato, tomato and bean.The insect digestive enzymes (proteases and a-amylases) from several herbivores and stored grain pests will be analyzed. We intend to investigate biochemical specificity of inhibitors and enzymes, their interaction, and changes during the inductionprocess in plants and insects. The effects of ingested inhibitors on the insects will be described in terms of biological parameters (e.g. growth rate, mortality, fecundity). The inhibitors will be supplied in the diet, or their levels will be modulated

    Úloha inhibitorů trávících enzymů hmyzu v obranyschopnosti rostlin vůči škůdcům

    V přirozené obranyschopnosti rostlin proti škůdcům hrají významnou úlohu rostlinné inhibitory trávicích enzymů hmyzu. Spektrum inhibitorů i enzymů se mění během komplexního procesu interakce rostliny s hmyzem (inhibitory indukované poraněním rostliny, enzymy indukované přítomností inhibitorů). Proteázové a a-amylázové inhibitory izolované z tkání bramboru, rajčete a fazole budou biochemicky charakterizovány a bude studován jejich inhibiční potenciál v testech in vivo a in vitro. Hmyzí trávicí enzymy (proteázy a a-amylázy) budou analyzovány u vybraných druhů herbivorů a skladištních škůdců. Výzkum bude zaměřen na biochemickou specificitu inhibitorů a enzymů, jejich interakci, a změny jejich složení během indukčních procesů u rostliny a hmyzu. Účinky inhibitorů požitých hmyzem budou sledovány na biologických parametrech (např. rychlost růstu, mortalita, plodnost). Hladiny inhibitorů v potravě hmyzu budou řízeny obohacením potravy isolovanými inhibitory nebo ošetřením živných rostlin chemickými
