The role of chloroplasts in mechanisms involved in replication and translation of viral-RNA in connection with host resistance

Project goals

The aim of project is the study and the characterization of role of chloroplasts in mechanisms of replication and/or translation viral-RNA in connection with susceptibility, tolerance and resistance of host plant and study of role of native inhibitors (coat protein CP, PR-proteins, antiviral factor AVF) of virus multiplication ("systemic acquired resistance"). Therefore we plan in this project in frame of resistance principle a) to investigate the relation between enhanced biosynthesis of viral-RNA precursors in chloroplasts and replication of viral-RNA and its coat protein and b) to elucidate the influence of natural inhibitors of viral multiplication (CP, PR-proteins, AVF). The results from this parts should be completed with the determination ofcontents of rRNA, of host and viral ss- and/or ds-RNA, of viral-CP, of PVY (potato virus Y) and TMV (tobacco mosaic virus) particles, PR-proteins and AVF (and their subcellular localisation) as well as with the study of intensities and regulation of

    Cílem projektu je studium a charakterisace role chloroplastů v mechanismech replikace a translace virové RNA u susceptibilních, tolerantních či resistentních rostlinných hostitelů a studium role přirozených inhibitorů reprodukce virů (obalového proteinuviru CP, PR-proteinů, antivirového faktoru AVF) ("systemic acquired resistance"). Proto bychom v tomto grantu chtěli v rámci studia principů resistence u susceptibilních a resistentních kultivarů tabáku řešit otázku vztahů mezi zvýšenou biosyntézou prekur?sorů virové RNA v chloroplastech a replikací vlastní RNA viru vč. tvorby jeho obalového proteinu a b) řešit otázku vlivu přirozených inhibitorů multiplikace virů (CP, PR-proteiny a AVF). Výzkum by spočíval ve stanovení obsahů rRNA, hostitelských avirových ss- a ds-RNA, virových CP, částic PVY (potato virus Y) a TMV (tobacco mosaic virus), PR-proteinů a AVF (a jejich subcelulární loka?lisace) současně se studiem intensit a regulačních mechanismů metabolických cest vedoucích k biosyntéze virové RNA
