Regulation of secondary metabolites biosynthesis in cell culture of Schisandra chinensis

Project goals

The intent of this project is the improvement of biotechnological methods designated for production of secondary metabolites with antioxidant activity, lignans and polyphenols, by plant cell cultures in vitro. The main objective of this study is the effect of precursors, elicitors and adsorbents on the production of antioxidant secondary metabolites by selected suspension cultures of Schisandra chinensis. It will be explored the impact of addition of different adsorbents, above all polymeric resins,on releasing of secondary metabolites from the cultured cells to media, on in situ adsorption of metabolites, on the overall increasing of its production and on possibility of continuous production of metabolites without harvesting the cells. The combined effects of precursors, elicitors and adsorbent will be also investigated as well as the biological activities of isolated secondary metabolites.     Regulace biosyntézy sekundarních metabolitů v buněčné kultuře Schisandra chinensis

    Záměrem tohoto projektu je zlepšení biotechnologických metod určených k produkci sekundárních metabolitů s antioxidační aktivitou, lignanů a polyfenolů, rostlinnými buněčnými liniemi in vitro. Hlavním cílem této studie je vliv prekurzorů, elicitorů a adsorbentů na produkci antioxidačních sekundárních metabolitů vybranými liniemi suzpenzních kultur Schisandra chinensis. Bude studován vliv přídavku různých adsorbentů, především polymerních pryskyřic, na uvolnění sekundárních metabolitů z kultivovaných buněk do media, na adsorpci metabolitů, na celkové zvýšení jejich produkce a na možnost kontinuální produkce metabolitů bez destrukce buněk. Společný vliv prekurzorů, elicitorů a adsorbentů bude rovněž studován stejně jako biologická aktivita izolovanýchsekundárních metabolitů. Studium obranných mechanismů rostlin, spojených s produkcí sekundárních metabolitů přispěje k optimalizaci kultivačních metod.
