Sequencing and the genome analysis of the strawberry vein banding virus

Project goals

The project will bring original knowledge about the sequence and genome arrangement of the Strawberry vein banding virus (SVBV), which is a new virus recently found in the Europe. The virus will be isolated from infected strawberry plants, where the presence of SVBV will be confirmed by S-blotting. Viral nucleic acid will be isolated and fragmented by restriction endonucleases, cloned into plasmid following standard producers and sequenced. Sequence and the genome arrangement of the SVBV will be analysed and compared with other caulimoviruses. Its sequence will be screened and specific primers for routine diagnosis of the virus by PCR will be prepared. Promoter directed the synthesis of the whole virus genome transcript will be selected from the bank of clones and used in artificial construction to control the expression of ß-glucuronidase (GUS) marker gene.

    Sekvenování a analýza genomu viru lemování žilek jahodníku

    Projekt má přinést nové poznatky o sekvenci, uspořádání genomu a jeho regulačních elementů v Evropě nově popsaného viru lemování žilek jahodníku (Strawberry vein banding virus). Virus bude izolován z infikovaných rostlin jahodníku, ve kterých bude přítomnost SVBV potvrzena S-blottingem. Virová DNA bude standardními metodami fragmentována, klonována do pBS plasmidu a sekvenována. Získaná sekvence bude analyzována, porovnána se sekvencemi jiných caulimovirů a budou podle ní připraveny specifické púrimérypro rutinní diagnostiku viru metodou PCR. Z vytvořené banky klonů bude vybrán klon obsahující virový promotor řídící syntézu genomové mRNA. Tento promotor bude upraven restrikčním štěpením a začleněn do plasmidu, kde bude řídit expresi signálního genu beta-glucuronidázy (GUS).
