Embryonic gene expression and posttranscriptional gene silencing during bovine preimplantation development
Project goals
Nowadays our knowledge of early mammalian embryogenesis is mainly based on studies performed on mouse model. However, in important aspects as activation of the embryonic genome, reprogramming, requirements on composition of cultivation medium, preimplantation bovines embryo is much more humanlike and alike to other mammals. There have been published a variety of studies describing a transcriptional profile of bovines embryo depending on different cultivation conditions. Especially when microarrays areused, we can obtained number of information on individual genes, but whose function in the early embryonic development is not known. We suggest using the results from our previous study, in which we found genes being expressed during genome activation using subtractive hybridization method. We will make posttranscriptional silencing of selected genes using double-stranded RNA (dsRNA) and find out whether there occurs retardation of the development. Comparison of transcriptional profile of silenced andcontrol embryos on microarrays will identify downstream genes of silenced genes. The genes with proved function can be subsequently used for characterisation of normal development of livestock species and a human being in in vitro embryo cultivation, for characterisation of embryonic stem, eventually somatic stem cells.
