    Mechanosensation in the Development and Healing of the Junctional Epithelium

    The Junctional Epithelium(JE) is a critical structure that separates the hostile environment of the oral cavity from the periodontium. Its malfunction leads to periodontitis, ultimately resulting in tooth loss. Mechanisms that activate the regenerative potential of the JE remain largely unknown. This collaborative project between Masaryk Uni (CZ) and Leipzig Uni (DE) aims to uncover the mechanosensory properties of the JE in the context of its development, mechanical loading, and inflammation. To achieve our goals, we will employ a comprehensive methodological approach, including metabolic labeling, targeted knockouts of PIEZO1/2, lineage tracing, spatial transcriptomics, and whole-mount 3D imaging. Ultimately, we will investigate the cellular and molecular mechanisms underlying de novo formation of JE around dental implants. This research will elucidate fundamental mechanisms of JE regeneration with relation to mechanosensation. Our findings may lead to the development of new therapeutic approaches in orthodontics, periodontal disease treatment or better dental implants integration.
