    Sensory cilia are antenna-like organelles of eukaryotic cells that detect stimuli and transmit signals, shaping gene expression and cell behaviour. Separated from the rest of the cell by a diffusion barrier, cilia have a distinct environment rich in membrane proteins and signalling enzymes. This heterogeneity is maintained by intraflagellar transport (IFT), which is essential for the entry, exit, and distribution of ciliary proteins, with disruptions leading to ciliopathies. The mechanisms by which IFT regulates protein entry and exit to maintain ciliary compartmentalization, as well as how disruptions in protein composition arise during ciliopathies or environmental changes, remain unclear. In my research, we will develop novel single-molecule fluorescence microscopy tools to resolve intracellular dynamics in sensory cilia of living C. elegans to address key gaps in the field. We will decipher how IFT preserves and regulates the diverse protein composition and distribution within cilia of living C. elegans, vital for sensory function at both cellular and organismal level.
