    The development of a novel electrospun fiber-based SERS nanotag platform designed for the multiplex detection of melanoma biomarkers is proposed. PLA electrospun fibers serve as solid supports for plasmonic nanoparticles, maximizing hotspot generation. The nanosystem is functionalized with 4-MBA and encapsulated in PEG/silica shells, enhancing stability against biochemical degradation and enabling bio-conjugation. Antibodies targeting three important melanoma biomarkers- CSPG4, Melan-A, and S100A8 are conjugated to the encapsulated NPs, enabling specific recognition and multiplex detection. The platform is validated through ultrasensitive detection of biomarkers at clinically relevant concentrations using SERS and in vitro studies with A375 melanoma cell lines. Bioimaging via confocal laser scanning microscopy will further assess biomarker interactions in normal skin cell lines, validating the platform’s applicability in complex biological matrices. This study holds promise as a novel, ultrasensitive, and cost-effective solution for early cancer diagnosis and monitoring.
